Microscopy Homework 1
1) The condenser lens concentrates light and makes illumination of image more uniform.

2) The bending of light is also known as refraction.

3) Total magnification = ocular lens magnification x objective lens magnification.

4) The total magnification of an image is 1200x and the magnification of the ocular lens is 10x. What is the magnification of the objective lens?

Y = objective lens
1200x = 10x X y
1200x / 10x = y y = 120x
Objective lens magnification = 120x

5) The limit of resolution is…

The limit of resolution is the closest two points that can be together for the microscope lens to make them appear separate; two points closer than the limit of resolution will blur together

6) You should always carry your microscope using two hands. One on the base of the microscope and the other supporting the neck.

7) At what objective is it proper to use oil immersion?

Only on objective 100x never on objective 10x or 40x

8) What are the proper two items to clean the microscope lens? And when do you clean it?

To properly clean the microscope you clean it with lens cleaner and lens paper so it does not scratch the lenses. You clean off all lenses including ocular lenses. Be sure not get the oil on the 10x or 40x objective. Lower stage and nose piece and set to 10x objective. Put dust cover on and return to cubby.

9) Number the following choices in chronological order. (use lab manual)
13 Work from lowest magnification to highest magnification – building up to the oil immersion magnification. Apply a conservative amount of oil on your slide. USE ONLY FINE ADJUSTMENT KNOB AND DO NOT TAKE YOUR EYES FROM THE MICROSCOPE WHILE YOU ARE ADJUSTING!!
5 Place a slide on the stage in the mechanical slide holder and center the specimen over the opening in the stage.
10 Scan for/examine specimen
4 Move scanning objective or low power objective in place. Do not rotate the nosepiece by objectives as this can CAUSE DAMAGE and cause them to unscrew from the nosepiece.

    The cover slip must be up if there is one. High magnification objective lenses can't focus through a thick glass slide; they must be brought close to the specimen, which is why coverslips are so thin. The stage may be equipped with simple clips (less expensive microscopes), or with some type of slide holder. The slide may require manual positioning, or there may be a mechanical stage (preferred) that allows precise positioning without touching the slide.…

    To focus a microscope using the high power lens, you have to turn the microscope knob to use the scanning lens. You have to be very careful for this next step; rotate the high powered lens, be sure to do it slowly and do not over turn the knob, if you turn the knob too much, the object you are viewing on the slide might crack, and if it’s a glass on, you can easily get cut.…

    Also, adjust the eyepiece by pushing close or separating them to the appropriate position. Perform the same process by changing the degree of the objective lens and focus to be able to see the grid clearly. Using Egeria densa, cut a leaf using a scissor and place it onto the slide glass using a forcep and using the droppers drop some water and place coverslip on top of the leaf by sliding the coverslip when placing on top to prevent air which produces bubbles. Place the slide glass on the stage, by moving the stage clip and move the coarse adjustment knob toward the experimenter to set the stage to the lowest. Use the 4x or 10x objective lens, so move the revolver to that and also the iris diaphragm lever matched to the objective lens.…
