A more stable structure is formed when the charge on the oxygen moves from being a lone pair to forming a double bond and the ether substituents are removed. In the presence of Hydronium, the negatively charged oxygen shares electrons with a hydrogen from the reagents. This mechanism allows for the formation of Salicylic Acid. When Acetic Anhydride is used as a reagent, the double bond on the ketone is transferred as a charge to the oxygen causing the opposite charges to attract. Once again, the electrophilic oxygen takes the lone pairs between the O-H bonds to form the final product of Aspirin and acetic acid. These mechanisms for this experiment can be seen in Scheme 1. Aspirin is classified as a Non-Steroidal Anti-Inflammatory Drug that is indicated for heart attack, pain, and fever.

Procedures Part A: Ten milliliters of 6M Sodium Hydroxide was added to two milliliters of methyl salicylate in a test tube. A white, crystal-like precipitate was immediately formed. Even upon shaking, the solution held its structure. The test tube was placed in a water bath containing boiling chips for while swirling the contents occasionally. The test tube was removed after 20 minutes. The precipitate no longer remained; the solution was now a pink liquid. The contents of the test tube were poured into a beaker to cool for approximately five minutes. Once cooled, 25 milliliters of 6M Hydrochloric Acid were slowly poured into the beaker.

    The goal of this experiment was to synthesize aspirin. In this experiment aspirin, also known as acetylsalicylic acid, was synthesized from salicylic acid and acetic anhydride. In the reaction the hydroxyl group on the benzene ring in salicylic acid reacted with acetic anhydride to form an ester functional group. This method of forming acetylsalicylic acid is an esterification reaction. Since this esterification reaction is not spontaneous, sulfuric acid was used as a catalyst to initiate the reaction. After the reaction was complete some unreacted acetic anhydride and salicylic acid was still be present in the solution as well as some sulfuric acid, aspirin, and acetic acid. Crystallization, which uses the principle of solubility, was then used to remove most of the impurities and caused the acetylsalicylic acid to precipitate out of the solution. Next, the crude product was then purified by adding water which further lowered the solubility of acetylsalicylic acid and dissolved some impurities from the crystal of aspirin. The aspirin was then isolated from the solution using a vacuum filtration apparatus. The percent yield of crude aspirin product was 91.89%. To purify the crude aspirin product a recrystallization procedure was performed. The percent yield of the purified aspirin product was 5.77%. Next a phenol test was performed on the crude aspirin product, the purified aspirin product, and salicylic acid as a control. The phenol test was to test the purity of the aspirin product that was created during the experiment. The crude aspirin product and the purified aspirin product had no color changes and remained orange when mixed with the iron (III) chloride solution, which means that there were no phenol groups in both the crude aspirin product and the purified aspirin product. The lack of reaction with the iron (III) chloride revealed that both the crude aspirin product and the purified aspirin product were pure aspirin. The salicylic acid turned…

In this lab, you will perform an organic synthesis to make aspirin. Aspirin is the trade name for the molecule acetylsalcylic acid. The earliest known use of this molecule has been traced back to fifth century B.C. The Greek physician Hippocrates described an extract of willow tree bark, a bitter powder that could be used to reduce fevers. In 1829, salicin was isolated from willow bark and used as a pain reliever. Unfortunately, salicin was not very popular, since it was found to be very acidic and a stomach irritant. In 1897, a German chemist named Felix Hoffman was working for the Bayer chemical company. Hoffman was looking for a less acidic pain reliever that his father could take for his arthritis. His research led to the synthesis of acetylsalicylic acid or aspirin. Bayer patented the name and marketed the product in 1899. It was a huge success and sales grew rapidly. In fact, the company set up by Friedrich Bayer & Company is generally reckoned to have been the first pharmaceutical company, and the production of aspirin is generally accepted to have laid the foundation of the modern pharmaceutical industry.
