ACTIVITY1 Assessing Starch Digestion by Salivary Amylase
1. List the substrate and the subunit product of amylase. _starch & maltose respectively__

2. What effect did boiling and freezing have on enzyme activity? Why? How well did the results compare with your prediction?__Boiling caused amylase to be denatured, thus inactivating the enzyme. Freezing has no effect. The function of an enzyme is directly related to its environment, like temperature.__

3. At what pH was the amylase most active? Describe the significance of this result. __ pH 7.0. Amylase is most active in neutral areas, such as the mouth and the small intestine (duodenum).__

4. Briefly describe the need for controls and give an example used in this activity. __Controls are needed to validate the results of the experiment. One example is testing for sugar using Benedict's test on Amylase The negative control was used to detect if amylase was contaminated with maltose or not.__

5. Describe the significance of using a 37°C incubation temperature to test salivary amylase activity. __37°C simulates an environment of normal body temperature. ___
A C T I V I T Y 2 Exploring Amylase Substrate Specificity
1. Describe why the results in tube 1 and tube 2 are the same. _In tube #1 the amylase is hydrolyzing the starch to glucose, & in tube #2 the glucose is already present in the hydrolyzed form.__

2. Describe the result in tube 3. How well did the results compare with your prediction? _ Amylase cannot digest cellulose, so tube #3 is not positive for Benedict’s test.__

3. Describe the usual substrate for peptidase. _A protein (BAPNA in this experiment).__

4. Explain how bacteria can aid in digestion. _Bacteria can aid in digestion by breaking down cellulose with the enzyme cellulase, humans do not produce this enzyme._

    This lab was focused on determining the optimal temperature of the enzyme amylase responsible for catabolizing starch polymers and to see how different temperatures affected the rate as well as how effectively the enzyme worked. To proceed with the experiment the group set up four different test tubes for each, bacteria and fungal amylase, and labeled them accordingly with different temperatures as well as different solutions . Then the spot plates were placed on the time and temperature table created with napkins and iodine was added to the first row were the solutions would be added later according to the time and temperature of each row. Because iodine reacts and turns a dark black color when starch is present they could determine the optimal temperature of each type of amylase by looking at and comparing the color changes. The group could reach a conclusion because they observed that at low temperatures more starch was present as well as at high temperature which was were the most starch was present. Because of these observations they concluded that the optimal temperature for amylase should be at about fifty-five degree celsius.…

    We then repeated this in two more test tubes. We put one of the test tubes in ice (0 C) we put another in the water bath (42 C) then we put one in boiling water (100 C). We repeated the same thing for the boiled amylase. We then checked at five minutes to see if the starch had been digested by the amylase. We did this by putting a little of each of the six samples into a spotting tile. We then added iodine solution to this to see if starch was present or not. I could tell if starch was present or not because if the solution went orange it meant that there was no starch present, so all the starch had been digested, if the solution went blue/black it meant that there was starch present, so the starch had not been fully digested. We recorded the results in a table. We then waited another two minutes so that all together the starch solution had been in there seven minutes.     The aim of this EEI was to test the effects of temperature on the activity of the enzyme Amylase. Solutions of starch and amylase were held at selected temperatures by various methods of temperature control. Once the solutions reached and maintained the desired temperature they were combined. Samples at timed intervals were then taken and reacted with a reagent to determine the effect the selected temperatures had on the reaction rate of enzyme and substrate. Results indicated that the enzyme functions efficiently at its optimum temperature (50oC) digesting the starch present and that any sign of enzyme function at 70oC is completely nonexistent.
